I'm thinking of replacing the standard Gotoh tuners that are on a guitar I built (16:1 with large Schaller-type button) with 510s (21:1 Contours).

A couple questions.

I'd like to swap the knobs between the two - can this be done?

Do the screw holes for these tuners line up on the back of the headstock? I'd rather not have to fill and redrill new holes.

Depends which Gotoh you started out with .. they arent all the same hole position (there are 510s that will direct replace the 301s if the hole position is like a grover - the schaller ones are different I believe - 381s maybe) .... I believe the buttons are interchangable however ..

I don't know for sure if the knobs will interchange. I did not know the contours were available in 21:1 I thought only the Delta's were 21:1.

Well I looked it up Delta's (21:1 gear ratio) come in two cases sizes. Contour case and mini contour case. So you are in a sense right they are contour's Delta contours to be explicit.

The mini Deltas are not 21:1 .. they are 18:1 in the catalog I have (SGS510Z) ... only full size Deltas (SGL510Z) are 21:1. The regular mini 510s (SGS510) are 15:1, regular full size 510s (SGL510) are 18:1. If you are looking to replace Grovers .. then you want the SGV version, in either std or Z type.

Deltas are a type of 510 tuner (510Z) .. so ANY of the 510 buttons can be had for them - there are 13 (plus one they dont even show - I know because I have them) options for the SGL and 7 for the SGS.
